Summary: Lettuce is a herbaceous plant, being considered the leafy vegetable most
consumed in Brazil. Commonly used for scientific studies, due to the sensitivity of the
species and the rapid germination. Plant Tissue Culture is an important tool for the
propagation of various kinds, however, the culture medium used may be costly. Aimed
to evaluate the development of Lactuca sativa (cv. Elba) comparing the MS culture
medium with a commercial formulation with and without vitamin. We used commercial
seeds with 99.5% purity and 74% germination rate, which, after sterilized, they were
inoculated in test tubes with 10 ml of culture medium containing the following
treatments: MS medium with vitamin (T1), MS medium without vitamin (T2) commercial
means with vitamin (T3) commercial means without vitamin (T4). Each treatment
comprised five replicates with 26 test tubes, totaling 130 seeds per treatment. The IVG
was carried out scores from the 4th day to the 7th day of germination. They were
considered germinated the seeds that had 2 mm of root protrusion in accordance with
recommendations of the Seed Analysis Rule. Data were evaluated according to
analysis of variance with a completely randomized design, forming a 2x2 factorial
design using the GENES program (Cruz, 2001). Tukey's test for multiple comparison of
averages was used.
